Due to me being lazy and not getting another tire for my spare, I've been driving around without a spare for a while (just a can of fix a flat). But I'm going to be driving probably around 400miles Saturday and I don't want to be without a spare for the trip. So right now I have one of my stock GT wheels in the trunk, but I know the stock wheel is heavier than the spare is. My question is, will it make a noticeable difference in gas mileage having the stock wheel in the trunk all the time? (thinking about just using that as a spare permanetely. Or at least for a while). And will it add stress to the suspension in the rear?

I doubt it would affect anything much, it's only 20 or so pounds heavier at the most.
